Probing interneuronal cell communication via optogenetic stimulation

نویسندگان

چکیده

This study uses an all-optical approach to probe interneuronal communication between spiral ganglion neurons (SGNs) and of other functional units, in this case cortex (CNs) hippocampus (HNs), for the first time. We combined a channelrhodopsin variant (CheRiff) with red genetically encoded calcium indicator (jRCaMP1a), enabling simultaneous optical stimulation recording from spatially separated small neuronal populations. Stimulation SGNs was possible both optogenetic manipulated HNs CNs, respectively. Furthermore, dependency on pulse duration stimulating light regard evoked response also observed. Our results pave way enable innovative technologies based “biohybrid” systems utilizing interaction different biological (eg, neural) systems. can improved treatment neurological sensorineural disorders such as hearing loss.

Optogenetic probing of mitochondrial damage responses.

It is now possible to functionally impair mitochondria through light illumination with high specificity. These optogenetic tools permit precise control on the timing, location, and extent of mitochondrial damage within a cell population with subcellular resolution, allowing quantitative probing of the various types of mitochondrial damage responses within cells. This approach can generally be e...

Optogenetic stimulation of neuronal repair

Environmental insult, disease or trauma can affect the physical integrity of neuronal circuits, and the inability of many neurons to regenerate injured axons invariably leads to irreversible neural dysfunction. The conserved second messenger cyclic adenosine monophosphate (cAMP) can promote axonal re-growth. Widely used pharmacological or genetic approaches to increase intracellular levels of c...

Probing eukaryotic cell mechanics via mesoscopic simulations

Cell mechanics has proven to be important in many biological processes. Although there is a number of experimental techniques which allow us to study mechanical properties of cell, there is still a lack of understanding of the role each sub-cellular component plays during cell deformations. We present a new mesoscopic particle-based eukaryotic cell model which explicitly describes cell membrane...

Optogenetic stimulation of the auditory pathway.

Auditory prostheses can partially restore speech comprehension when hearing fails. Sound coding with current prostheses is based on electrical stimulation of auditory neurons and has limited frequency resolution due to broad current spread within the cochlea. In contrast, optical stimulation can be spatially confined, which may improve frequency resolution. Here, we used animal models to charac...

Exosomes as a novel way of interneuronal communication.

Exosomes are small extracellular vesicles which stem from endosomes fusing with the plasma membrane; they contain lipids, proteins and RNAs that are able to modify receiving cells. Functioning of the brain relies on synapses, and certain patterns of synaptic activity can change the strength of responses at sparse groups of synapses, to modulate circuits underlying associations and memory. These...
